1. Porcine Parvovirus Antibody Diagnostic ELISA Kit Usage
GreenSpring® Porcine Parvovirus (PPV) ELISA Kit is used for the
qualitative detection of Porcine Parvovirus (PPV) IgG antibodies in
pig serum; the assessment of the immune conditions of Porcine
Parvovirus in pig farms and the epidemiological investigation of
Porcine Parvovirus.
2. Principle
GreenSpring® Porcine Parvovirus (PPV) Antibody ELISA Detection Kit
is made from antigen-coated microtiter plates (PPV antigen-coated)
and other reagents. The principle of solid-phase ELISA was applied
to PPV-Ab in serum, and then the enzyme conjugate was added to the
microplate to specifically bind to the complex of coated antigen +
PPV-Ab + enzyme-labeled anti-pig IgG. With TMB substrate, it
produces a certain amount of color. The depth of color is related
to the content of PPV-Ab. When the color value is greater than the
critical value, the pigs are well inoculated or have natural
infection.

4. User Notices and Warnings
1. This kit is for research use only.
2. Please read the manual carefully before use.
3. The experimental garbage should be sterilized by high pressure
steam at 121℃ for 30 minutes, or treated with 5.0g/L sodium
hypochlorite disinfectant for 30 minutes and then discarded.
4. Microplates removed from refrigeration should be equilibrated,
dried at room temperature, and ready to be opened. Return unused
MicroWell plates to dry foil bags and seal at 4 °C. Unused liquid
reagents should be covered and stored together with other
components at 2-8°C away from light.
5. Samples and reagents should be added using a micropipette and
frequently demonstrated for accuracy.
6. When adding washing buffer, it should be topped up but not
overflowed to avoid free enzyme or cross-contamination between
wells.
7. The stop solution is corrosive, immediately rinse with plenty of
water when it comes into contact with skin or clothes.

Epidemiological investigation
The dead fetus, piglets and excreta in the uterus of infected sows
contain high titer of virus, while the live pigs produced by
infected pigs may carry virus for a long time or even a lifetime.
The infected boars are also the most dangerous source of infection
of the disease. The virus can be isolated from the semen, spermatic
cord, epididymis and gonads of boars. The boars can be transmitted
to susceptible sows through breeding and spread the disease.
Polluted pig houses are the main storage of porcine parvovirus. The
infected pigs can still be infected after the sick pigs are removed
from the empty pen for 4.5 months and thoroughly cleaned before
being put into the susceptible pigs. Contaminated food and pig
saliva can be infectious for a long time.
Piglets, embryos and fetuses are vertically infected by infected
sows; Boars, finishing pigs and sows are mainly infected by
contaminated feed and environment through respiratory tract,
reproductive tract or digestive tract; Most of the infection of
primiparous sows occurred during breeding with infected boars; Mice
can also spread the disease.
The disease is highly infectious. The introduction of the
susceptible healthy pig herd - Danvirus can cause almost 100%
infection in the pig herd within 3 months; The infected pigs
remained seropositive for a long time. This disease mostly occurs
in spring and summer festival or sow litter and mating season. When
sows are infected in the early pregnancy, the mortality of embryos
and fetal pigs can be as high as 80%~100%. Sows are most
susceptible to infection in the first 30 to 40 days of pregnancy.
Infection at different times of pregnancy will cause different
symptoms, such as stillbirth, abortion, mummy, weak piglets and
long term infertility of sows. The infection rate of virus is
proportional to the age of animals.
